A new variety of Bacopa monnieri obtained by in vitro polyploidization Electron. J. Biotechnol.

The commercial value of Bacopa monnieri, a widespread herbaceous plant in Argentina, can be substantially improved increasing its flower size by chromosome doubling with colchicine. MS supplemented with 0.25 mg/L of 6-benzylaminopurine proved to be an appropriate medium for the in vitro multiplication of nodal segments of B. monnieri. Polyploidization was achieved submerging nodal segments during 24 or 48 hrs in colchicine solution 0.001 and 0.01% P/V, in 1% DMSO. Segments submerged in water and in DMSO 1% aqueous solution were used as controls. DNA contents from recovered plants was measured and characterized and their phenotype described. Autumn crocus poisoning of cattle is characterized by severe diarrhea caused by alkaloid colchicine. Previously, we examined pathologically this poisoning in cattle and reported that enterotoxic lesions were closely associated with apoptosis. To examine enterotoxicity of autumn crocus more precisely, a reproductive study was performed using guinea pigs and mice, and pathological findings associated with autumn crocus poisoning were compared with those of colchicine. Each group of guinea pigs given the bulb of autumn crocus or colchicine exhibited severe diarrhea. Flow cytometry allows to estimate the DNA content of a large number of plants quickly. However, inadequate protocols can compromise the reliability of these estimates leading to variations in the values of DNA content the same species. The objective of this study was to propose an efficient protocol to estimate the DNA content of Brachiaria spp. genotypes with different ploidy levels using flow cytometry. ABSTRACTArtificial chromosome duplication is one of the most important process in the attainment of doubled haploids in maize. This study aimed to evaluate the induction ability of the inducer line KEMS in a tropical climate and test the efficiency of the R1-Navajo marker by flow cytometry to evaluate two chromosome duplication protocols and analyze the development of the doubled haploids in the field. Induction and identification of polyploids in Cattleya intermedia Lindl. (orchidaceae) by in vitro techniques Ciência Rural

The effect of colchicine on the protocorm-like bodies of Cattleya intermedia L. (Orchidaceae) cultured in vitro was studied. Different concentrations of the drug (0.00, 0.05, 0.10 and 0.20%) as well as two times of culture (4 or 8 days) were tested to determine the best treatment for the induction of tetraploid plants. Of the 3 colchicine levels applied, 0.05 and 0.10% of drug treatments appeared to be effective on the production of mixoploids and tetraploids and could be further used in breeding programs. The suitability of stomatal area and stomata density as criteria for the distinction between diploids and tetraploids was also tested. Peanut (Arachis hypogaea) is the fifth most produced oil crop worldwide. Besides lack of water, fungal diseases are the most limiting factors for the crop. Several species of Arachis are resistant to certain pests and diseases. This study aimed to successfully cross the A-genome with B-K-A genome wild species previously selected for fungal disease resistance, but that are still untested. We also aimed to polyplodize the amphihaploid chromosomes; cross the synthetic amphidiploids and A. hypogaea to introgress disease resistance genes into the cultivated peanut; and analyze pollen viability and morphological descriptors for all progenies and their parents.
